Histone H3k27 Trimethylation

Histone H3K27 trimethylation is a chemical tag that lives on the tail of a protein called histone H3, one of the core components around which DNA is wrapped. The tag consists of three small methyl groups attached to a specific amino‑acid building block known as lysine at position twenty‑seven. By adding this dense layer of methyl groups, cells create a surface that attracts other proteins designed to tighten the surrounding chromatin and keep nearby genes quiet.

The importance of this mark comes from its role as a stable signal for long‑term gene repression. When a cell wants to lock down a developmental program, silence one copy of a chromosome, or maintain the identity of a particular tissue, H3K27 trimethylation helps preserve that state through many rounds of division. Disruptions in the placement or removal of this modification are linked to a range of disorders, most notably several cancers where genes that should be silent become active, or vice versa.
